01 · What is Cagrilintide?

Cagrilintide (AM-833) is a modified amylin analogue engineered for once-weekly dosing in preclinical and clinical research. It is structurally related to the approved amylin analogue pramlintide but with substitutions that extend its circulating half-life to approximately one week.

02 · Mechanism of action

Cagrilintide activates amylin (AMY) and calcitonin receptors expressed in the area postrema and hypothalamic feeding circuits. In preclinical models the result is delayed gastric emptying, suppression of postprandial glucagon and a central satiety signal that is mechanistically distinct from — and additive to — GLP-1 receptor agonism. This is the rationale for the CagriSema combination programme.

03 · Handling, reconstitution and storage

Lyophilised Cagrilintide is stored at -20 °C, desiccated, protected from light. Once reconstituted with bacteriostatic water (0.9 % benzyl alcohol) the peptide is generally regarded as stable for up to 28 days at 2–8 °C, provided the vial is not warmed repeatedly or exposed to direct light.

Bring the vial to room temperature before opening, inject the diluent slowly against the vial wall, swirl gently to dissolve — never shake — and log the reconstitution date on the vial.
