Cagrilintide: the amylin analog behind CagriSema
Cagrilintide is a long-acting amylin analog dosed once weekly. Amylin is co-secreted with insulin from pancreatic beta cells and slows gastric emptying, suppresses glucagon, and increases satiety through a different central mechanism than GLP-1. The combination product — CagriSema — pairs cagrilintide with semaglutide 2.4 mg to target both pathways.
Phase 2 data suggested additive weight loss over semaglutide alone, though the magnitude of the incremental effect has been debated after mixed Phase 3 readouts in 2024–2025.
